6  Capture Antibody Preparation

This procedure prepares all the capture antibodies, to be ready for coupling them to the MagPlex beads and storing for future use.

6.3 Protocol

  1. Make up PBS as per the Buffers and Reagents protocol.
  2. Set out and label 10 microtubes per capture antibody.
  3. Remove antibodies from freezer storage.
  4. Add 100 µL of the PBS to each lyophilised antibody (anti IL-8 and anti MxA are already liquid at 1 mg/ml so do not need any liquid adding).
  5. Allow to rehydrate for a few minutes, vortex gently then spin liquid down briefly.
  6. Aliquot all antibodies into 10 µL volumes, or 50 µl volumes for IL-8.
  7. Store at -20°C or below.

All capture antibodies are now at the same stock concentration of 1 mg/mL.
